Dio is the son of Dario Brando, an alcoholic and abusive father. Back when Dio was just a boy, he was brought into the household of George Joestar after the mysterious death of his father, so as to repay the debt he owed Dio's father for saving his life. But soon Dio's personality caused trouble for George's young son, Jonathan Joestar. Even more of a sociopath than his father, Dio was resolute to shatter Jonathan's spirit and take his place as the favored son, even going so far as to arrange for Jonathan's dog Danny to be burned to death.

He is featured initially as the main antagonist of the series' first part, Phantom Blood , and later returns as the main antagonist of the series' third part, Stardust Crusaders , now solely referred to as Dio. The poor son of Dario Brando, an abusive alcoholic who worked Dio's mother to death, Dio first appears as a young misanthrope who, via the power of an ancient stone mask, becomes an immortal vampire. A century after the events of the first part, he resurfaces as the user of the time-stopping Stand named The World. As the series' most prolific villain, his defining trait is his staunch ambition, which develops into a " might makes right " philosophy and a drive for absolute control through absolute power. When beginning the series, Dio was the character that Araki looked forward to drawing the most.

When he was in junior high school, Koyasu loved movies so much that he would watch them every day without going to school, aspiring to become a film actor himself. A scene in Adieu Galaxy Express sparked his interest in voice acting. He entered a training school after graduating from high school. Koyasu knew about JoJo's Bizarre Adventure while it was being serialized though he doesn't recall if he actually knew the plot or only certain maniacal details about it. At the time it was serialized, he didn't think he would become a voice actor, and he also never thought he would be asked to play Dio Brando in a TV anime for it. However, when he heard that they were auditioning for Dio, he thought there would be no one else more suitable than him. He felt like if he had the chance to play the role, then he would risk his life to do so.
