Guanylyl cyclase

Guanylate cyclase Guanylyl cyclase 4. It is often part of the G protein signaling cascade that is activated by low intracellular calcium levels and inhibited by high intracellular calcium levels. Like cAMPcGMP is an important second messenger that internalizes the message carried by intercellular messengers such as peptide hormones and nitric oxide and can also function as an autocrine signal, guanylyl cyclase.

Nitric oxide, bicarbonate, natriuretic peptides ANP, BNP and CNP , guanylins, uroguanylins and guanylyl cyclase activating proteins GCAPs activate a family of enzymes variously called guanyl, guanylyl or guanylate cyclases that catalyze the conversion of guanosine triphosphate to cyclic guanosine monophosphate cGMP and pyrophosphate. Intracellular cyclic GMP is a second messenger that modulates: platelet aggregation, neurotransmission, sexual arousal, gut peristalsis, blood pressure, long bone growth, intestinal fluid secretion, lipolysis, phototransduction, cardiac hypertrophy and oocyte maturation. This review briefly discusses the discovery of cGMP and guanylyl cyclases, then nitric oxide, nitric oxide synthase and soluble guanylyl cyclase are described in slightly greater detail. Cyclic GMP was first purified and identified in rat urine in by Ashman and colleagues [ 1 ]. The enzymes that catalyze the conversion of GTP into cGMP were discovered 6 years later by three separate groups [ 2 — 4 ]. The cGMP synthesizing activity was initially called guanyl cyclase, then guanylate cyclase and in recent literature it is most often referred to as guanylyl cyclase. As a result of these initial studies, it was determined that the enzyme could be separated into distinct soluble and particular forms with unique kinetic properties and tissue distributions [ 5 , 6 ]. However, the true diversity of the family was not fully appreciated until molecular cloning identified cDNAs that encoded individual enzymes. The soluble forms exist as heterodimers and the transmembrane members are homodimers, except for GC-C, which appears to be a homotrimer. The minimal guanylyl cyclase catalytic unit is a dimer [ 8 , 9 ]. The soluble forms contain an amino terminal heme-binding, dimerization and carboxyl-terminal catalytic domains.

Mechanisms of ligand discrimination by heme proteins.

Nitric oxide NO , carbon monoxide CO , oxygen O 2 , hydrogen sulfide H 2 S are gaseous molecules that play important roles in the physiology and pathophysiology of eukaryotes. Various hemoproteins play a significant role in sensing and transducing cellular signals encoded by gaseous molecules or in transporting them. Soluble guanylyl cyclase sGC is a hemoprotein that plays vital roles in a wide range of physiological functions and combines the functions of gaseous sensor and signal transducer. Carbon monoxide CO , nitric oxide NO , oxygen O 2 and hydrogen sulfide H 2 S are gas molecules that are similar not only due to their small size, similar water solubility and membrane permeability but also the fact that they can be generated by different living organisms. In many cases these gaseous molecules are essential for cellular communications and act as signaling molecules that trigger various physiological responses Girvan and Munro, ; Liebl et al.

Jurasza St. For years, guanylate cyclase seemed to be homogenic and tissue nonspecific enzyme; however, in the last few years, in light of preclinical and clinical trials, it became an interesting target for pharmacological intervention.

Kinetics and equilibria of soluble guanylate cyclase ligation by CO: effect of YC CO is a well-known gaseous ligand for various hemoproteins. Nat Commun 12 , Categories : EC 4. The conformational changes in the sensor module and the transducer module induced by agonists, stimulators and activators are finally conveyed to the catalytic module via the inter-module connections to open the GTP substrate binding pocket. Here, we present cryo-electron microscopy structures of human sGC in complex with NO and sGC stimulators, YC-1 and riociguat, and also in complex with the activator cinaciguat. The importance of cysteine thiol in sGC activity and function is well recognized Beuve, These guanylyl cyclases probably evolved from adenylyl cyclase based upon topology [ 13 ]. Chondrocyte overexpression of CNP rescued growth in a mouse model of the most common type of human dwarfism [ 58 ]. Schrammel, A. Mutations in the genes that encode mGCs or pathologic activation of these enzymes or their activators are associated with diseases of the cardiovascular, skeletal, intestinal or visual systems. Some of these stimulators, such as riociguat and vericiguat have been approved for treatment of conditions of pulmonary hypertension and heart failure, respectively Ghofrani et al. Kubes, P. A recent cryo-EM map of M. Becker, E. Free Radic. NO activates sGC by binding to the sixth position of the heme ring, which breaks the bond between the axial histidine and iron to form a 5-coordinated ring with NO in the fifth position. However, when similar mutation was introduced to the full length sGC, no oxygen binding was observed, while the sGC mutant was fully functional Martin et al. NO binding to the heme causes the cleavage of the heme-His coordinate bond Figure 1C. The interactions of NO with sGC. Soluble guanylyl cyclase is recognized as the most sensitive physiologic receptor for nitric oxide.
