LoaP is a class of proteins that has the potential to induce bacterial species into creating natural products such as antibiotics. LoaP can be placed as an insert into bacterial genomes, where it can potentially drive gene expressions and subsequently activate secondary biosynthetic pathways. Here, a construct was cloned by placing LoaP as an insert into the plasmid of Clostridium beijerinckii. However, this LoaP overexpression strain failed to produce a compound that its wild-type creates. LoaP appeared to decrease production rather than increase production of certain metabolites, as the wild-type strain gave rise to more natural products than the transformant LoaP strain.

In this study, we analysed the phylogenetic distribution of the large, widespread NusG family of transcription elongation proteins and found that it includes a cohesive outgroup of paralogues herein coined LoaP , which are often positioned adjacent or within gene clusters for specialized metabolites. We established Bacillus amyloliquefaciens LoaP as a paradigm for this protein subgroup and showed that it regulated the transcriptional readthrough of termination sites located within two different antibiotic biosynthesis operons.

Background: Local adipose tissue AT cooling is used to manage obesity and overweight, but the mechanism is unclear. Results: We report that i single sessions of tissue cryotherapy lead to significant loss of tissue volume in the time frame of hours and ii multiple daily procedures lead to a cumulative decline in AT, as assessed by waist circumference, body weight, and BMI, confirmed by whole-body dual-energy X-ray absorptiometric scanning. In addition, iii blood analysis following tissue cryotherapy found no significant changes in biochemical parameters including markers of inflammation. Moreover, iv calculations of heat extracted and of compensatory weight loss taking place through thermogenesis are substantially consistent with the observed loss of AT.
