Nathaniel Essex was born in Victorian England during the 19th Century. He was obsessed with Charles Darwin's Theory of Evolution. Although he was genuinely passionate about his work, he also cared a great deal for his wife Rebecca Essex and their son. Disaster struck when their son died, aged four, of various birth defects. Essex started to intensify his work and theories about evolution to cope with the pain of this loss, ignoring his again pregnant wife as a result.

Created by writer Chris Claremont , the character was first mentioned as the employer behind the team of assassins known as the Marauders in The Uncanny X-Men December , and later seen in silhouette in The Uncanny X-Men , with both issues serving as chapters of the " Mutant Massacre " crossover. His appearance was designed by artist Marc Silvestri. A villain who usually prefers to act through agents and manipulation, Mr. Sinister is born Nathaniel Essex in Victorian London. As he learns about mutants superhuman beings born with the X-gene , Essex encounters the mutant villain Apocalypse. The two become allies and Apocalypse uses alien Celestial technology to transform the British scientist into Mr. Sinister, an ageless man with super-powers. In the modern-day, Sinister develops a great interest and protective attitude towards the mutant heroes Cyclops and Jean Grey , believing their DNA can create the ultimate mutant. This and other factors lead him to have repeated clashes with the X-Men a group Cyclops and Jean Grey helped found and related teams. Several co-existing Mr. Sinister clones all sharing the original's memories and basic personality later formed a community.

After experimenting with a cell sample from the latter, Sinister gains mr sinister control over his physical form, allowing regenerative abilities and shape-shifting.

Mister Sinister was an Apocalypse-genetically created mutant hellbent on creating the perfect X-gene clone. Nathaniel Essex was born at Milbury House on the outskirts of London. As a child, Nathaniel traveled to Greenwich Park, south of the River Thames in London, and was amazed by the endless number of people, walking what he believed to be preordained paths for them. Essex believed that the sanctity and purity of it was Heaven on Earth. He gained a full scholarship to Oxford University and later became a biologist in Victorian England. He developed advanced theories on human evolution and was obsessed with Charles Darwin's theory of evolution, though he felt Darwin and his contemporaries were shackled by too many moral constraints, and that science was beyond morality. Essex dreamed of bringing the perfection of the evolution of the human race to the masses.

Through the years, Mister Sinister became one of the greatest geneticists to have ever lived, skilled in cloning , creating new life, and even in the manipulation of mutant DNA for enhancing powers, incorporating mutant DNA to his own.

Using his stealth and tracking skills, Gambit leads the group to the Morlock community but abandons the group when he learns they intend murder. He can control the actions of others, and cannot be detected by any form of cameras, sensors, or anything but the naked eye. According to Magneto , Sinister would offer candy to children in exchange for blood samples. Sinister joining the new mutant community of the island Krakoa , and joining its ruling Quiet Council alongside Magneto , Professor X, Apocalypse, and others. At the time of their encounter, Logan had become feral after the events that had led to the death of his first love, Rose O'Hara. He hired a group of thugs, whom he named Marauders, to kidnap people off the streets of London so he could perform experiments on them; he even went so far as to dig up his dead son and experiment on him. When Apocalypse demands Sinister create a plague that will wipe out much of humanity, leaving only "the strong" alive, the scientist refuses, arguing that cruelty without purpose is ignorance and the enemy of science.
