Pancreatic acini

The pancreatic acinar cell is the functional unit of the exocrine pancreas. It synthesizes, stores, and secretes digestive enzymes. Under normal physiological conditions, digestive enzymes are activated only once they have reached the duodenum, pancreatic acini.

A pancreatic acinus is a functional unit of the exocrine pancreas producing digest enzymes. Its pathobiology is crucial to pancreatic diseases including pancreatitis and pancreatic cancer, which can initiate from pancreatic acini. However, research on pancreatic acini has been significantly hampered due to the difficulty of culturing normal acinar cells in vitro. In this study, an in vitro model of the normal acinus, named pancreatic acinus-on-chip PAC , is developed using reprogrammed pancreatic cancer cells. In this model, human pancreatic cancer cells, Panc-1, reprogrammed to revert to the normal state upon induction of PTF1a gene expression, are cultured.

Michelle M. Cooley 1 , Elaina K. Jones 1 , Fred S. Gorelick 2 , and Guy E. Groblewski 1. A key function of the exocrine pancreas is the production of digestive enzymes. The pancreatic acinar cell synthesizes, stores, and secretes the proenzymes and enzymes needed to digest dietary proteins, carbohydrates, and lipids. Meeting these functional requirements necessitates that the acinar cell has very high rates of protein synthesis and export. Nascent proteins undergo folding, select modifications, concentration, segregation from other classes of proteins, and vectorial movement before reaching their final destination in secretory zymogen granules. These are concentrated in the apical pole of the acinar cell. Eating stimulates neural and hormonal pathways that mediate acinar cell zymogen granule exocytosis into the pancreatic duct. The exocrine pancreas has two major physiologic functions: it supplies the proenzymes and enzymes needed for digesting dietary lipids, carbohydrates, and proteins; and secretes a bicarbonate-rich fluid that neutralizes acidic gastric secretions and thus provides the correct pH for intestinal digestion by pancreatic enzymes. The acinar cell has been a model system for foundational studies of protein synthesis and export. After electron microscopy was developed, for example, cell biologists first visualized organelles and established their function by studying acinar cells Figure 1. Here, we focus on acinar cell protein synthesis, trafficking, and processing in the pancreatic acinar cell necessary for its central role in producing digestive enzymes.

Human pancreatic exocrine cells were cultured in 3D suspension and formed pancreatospheres composed of acinar-derived and duct-like cells. We investigated, up to 6 days, the fate of human pancreatic acinar cells using fluorescein-conjugated Ulex Europaeus Agglutinin 1 lectin, a previously published acinar-specific non-genetic lineage tracing strategy.

Pancreatic islets, also called the islets of Langerhans, are regions of the pancreas that contain its hormone-producing endocrine cells. The pancreas serves two functions, endocrine and exocrine. The exocrine function of the pancreas is involved in digestion, and these associated structures are known as the pancreatic acini. The pancreatic acini are clusters of cells that produce digestive enzymes and secretions and make up the bulk of the pancreas. The endocrine function of the pancreas helps maintain blood glucose levels, and the structures involved are known as the pancreatic islets, or the islets of Langerhans. Pancreatic islets or islets of Langerhans : The islets of Langerhans are the regions of the pancreas that contain its endocrine hormone-producing cells. The pancreatic islets are small islands of cells that produce hormones that regulate blood glucose levels.

A role for SNAP29 in acinar cell function has not been described. It follows that molecular chaperones are critical for pancreatic acinar cell function. The aspirates consist primarily of cohesive fragments creating acini, cellular cords, or solid nests of neoplastic epithelium, composed of clusters of uniform cells with smoothly contoured, eccentric nuclei containing one or two prominent nuclei nucleoli[ 16 , 17 ]. A small population was double positive for CA Cytopathology of the pancreatobiliary tract-the agony, and sometimes, the ease of it. ER-resident chaperones assist newly translated polypeptides by recognizing and interacting with key protein regions and shielding them from outside interference while the necessary modifications are made. BMC Dev Biol. The features are becoming clearer. Clathrin coat formation at Golgi membrane is mediated by assembly of clathrin adaptor protein complexes AP1, AP3, and AP4; these multisubunit complexes associate with cytoplasmic domains of specific Golgi transmembrane proteins to nucleate clathrin coat assembly 71, ZGs have been shown to undergo exocytosis at the basolateral plasma membrane during acute pancreatitis For example, the initial acinar cell cytoplasmic calcium signal that mediates protein secretion depends on an ordered, spatial release from distinct ER stores. In these favorable pancreatic cancers, aggressive surgical resection with negative margins is associated with long-term survival.

The pancreas serves digestive and endocrine functions, and it is composed of two types of tissue: islets of Langerhans and acini. The pancreas is a glandular organ in the digestive system and endocrine system of vertebrates. It is both an endocrine gland that produces several important hormones—including insulin, glucagon, somatostatin, and pancreatic polypeptide—as well as a digestive organ that secretes pancreatic juice that contain digestive enzymes to assist the absorption of nutrients and digestion in the small intestine.

Sorting of cargo receptors and lysosomal transmembrane proteins requires sorting signals present in cytosolic domains which interact with components of clathrin coats or an adaptor protein complex. It was previously reported that human acinar cells in 2D monolayer culture spontaneously transdifferentiate into a duct-like phenotype, with acquisition of proliferative potential and induction of epithelial-mesenchymal transition EMT 14 , Holen et al [ 8 ] found that median survival for surgically resected cases was 36 mo, compared to 14 mo for those without surgical resection[ 8 ]. Lipase may be used as a tumor marker in these patients. The purpose of these distinct mechanisms of secretion is unknown, but they likely serve to both ensure that some digestive enzymes will be present in the small intestine at all times and provide a secretory response that is proportional to luminal nutrients. ACC supports the use of combination therapies based on gemcitabine or 5-fluorouracil and incorporating irinotecan, a platinum analog, or docetaxel in patients with advanced disease. The autophagy regulatory protein ATG9 is the only membrane-associated autophagy protein that resides in the ER. Nascent proteins are synthesized in about 5 min in the endoplasmic reticulum; then move by transport vesicles to the Golgi complex, where they exit at about 20 min in condensing vacuoles a. Dev Cell. Consequently, institutional and multi-institutional reports of ACC continue to be essential for performing a more comprehensive analysis of the presentation, pathology, natural history, and treatment-related outcomes of ACC Resection and chemotherapy 33 Resection and RT 10 Resection and chemoradiation Other treatment 1 Matos et al [ 75 ] 17 Resection 12 ACC requires aggressive surgical resection. Additional secretory pathways identified in acinar cells. Even though it has not yet been proven in major clinical trials, preliminary findings indicate that tumors with BRAF fusions react to MEK inhibitors, providing ACC with targeted therapy[ 41 - 43 ]. Furthermore, the acinar-derived cells, i.
